Introduction:
The developmental process of the nervous system is regulated by many genes, among which the Ninj2 gene is a gene that encodes a neuron-specific protein. In previous studies, the Ninj2 gene has been found to play an important role in the development of the nervous system. However, the specific mechanisms of action and regulatory pathways of the Ninj2 gene remain unclear. Therefore, in this study, we investigated the role of Ninj2 gene in nervous system development by performing knockout experiments and further explored its regulatory pathways.
Materials and Methods:
In this study, mice were used as the study subjects to knock down the Ninj2 gene by CRISPR/Cas9 technology. The role of Ninj2 gene in the development of the nervous system was explored by histological and behavioral analysis of knockout mice. Meanwhile, the regulatory role of Ninj2 gene on Notch signaling pathway was analyzed by Western blot and real-time fluorescence quantitative PCR.
Results:
Knockdown of the Ninj2 gene resulted in abnormal development of the nervous system in mice, as evidenced by reduced neuron numbers, abnormal synapse formation and behavioral defects. In addition, we found that the Ninj2 gene can affect neurological development by regulating the Notch signaling pathway. Specifically, knockdown of the Ninj2 gene leads to activation of the Notch signaling pathway, which affects neuronal differentiation and synapse formation.
Discussion:
The results of this study suggest that the Ninj2 gene has an important role in the development of the nervous system. Through knockdown experiments, we found that the Ninj2 gene can affect key processes such as neuron number and synapse formation, thus influencing the behavioral performance of mice. In addition, we also found that the Ninj2 gene can affect neurological development by regulating the Notch signaling pathway. These results provide new ideas and directions for an in-depth study of neurological development.
Conclusion:
This study confirms the important role of the Ninj2 gene in the development of the nervous system and provides new ideas and directions for the in-depth study of the development of the nervous system. In addition, we also found that the Ninj2 gene can affect nervous system development by regulating the Notch signaling pathway, which provides important clues for future studies.
