Protamine-mediated efficient transcellular and transmucosal delivery of proteins

For the treatment of patients with chronic diseases, protein and peptide drugs are generally administered via parenteral route, but the parenteral route suffers from inconvenience and poor compliance due to the poor membrane permeability and low stability of these drugs in the gastrointestinal tract. To overcome this challenge, the Wood study reported a non-parenteral drug delivery method by physical mixing with fisetin (an FDA-approved peptide), which was shown to promote tubularization and rearrangement of cellular actin and enhance intracellular protein delivery compared to polyarginine 8 (R8, cell-penetrating peptides (CPPs)), and in mouse experiments, fisetin was shown to overcome the mucosal and In mouse experiments, fisetin was shown to overcome the mucosal and epithelial barriers, modulate adherens junctions, and facilitate insulin penetration into the lamina propria for systemic uptake. The non-parenteral (intranasal) delivery of fisetin developed by the team has potential utility in the treatment of diabetes in humans.
